Biochemical and molecular markers – mechanisms, major types, typical examples of their use in taxonomy, in-vitro cultures, somaclonal variation. Protein markers and Isozymes. Polyacrylamide gel (PAGE) electrophoresis. Specific and non-specific protein staining. Chemotaxonomy – principles, major groups of compounds used, methods of their analysis (gas and liquid chromatography, mass spectrometry), data analysis. Development and characterization of molecular markers. Genetic and Physical mapping; mapping populations. Molecular Cytogenetics. Methods of physical positioning of DNA sequences on chromosomes and interphase nuclei. Utilization of methods based on hybridization of DNA molecules and polymerase chain reaction (PCR) as sources of molecular markers and data analysis. Molecular markers as a tool for clone development and gene characterization.
